Fish is one of the sources of protein and it has numerous bacteria and disease associated with it. The objective of the study is to determine the presence of resistant gene in bacteria associated with Clarias gariepinus. The gills, skin and the intestines of sixteen C. gariepinus of about 5g each were taken from different fish pond in Ijebu-Ode, the fish samples were dissected with surgical blade and the samples were inoculated on Samonella-shigella agar, Eosinemethylene blue agar. Antimicrobial susceptibility tests was done using Bauer Kirby disc diffusion method on Mueller Hinton agar. The pure colony was identified and the bacteria strains were inoculated in Nutrient broth overnight for DNA extraction. PCR analysis was done using tetracycline resistance gene marker. The morphometric characteristics shows that Total length (cm) (26.88±4.05) and Standard length (cm) (22.75±3.92), body weight (g) (128.81±20.89). There were five types of bacteria strains that were discovered present in the samples taken from C. gariepinus this includes Escherichia coli, Shigella, Samonella, Proteus species and Klebsiela species. Gills has the highest number of bacteria followed by skin and intestine has the least number of bacteria. Salmonella have the highest strain frequency while Shigella with the low frequency. The sensitivity test shows that the bacteria are sensitive to Gentamycin, Tarivid and Ciprofloxacin while few are resistant to other antibiotics. Molecular detection of Tetracycline (A) resistance gene shows that all the isolates obtained were resistant to tetracycline antibiotics.
