The rate of consumption of local indigenous fruit like Gambeya albida as mode of vitamins for the rural dwellers, such as children and women in African nations is alarming. This species provided necessary vitamin supplement for proper growth of the African people. The previous research was focus on the genetic variation in Osun State, while this research focus on genetic variation of Southwestern Nigeria. The genetic variation of Gambeya albida is very important to determine the main sources of this species for the improvement of this species. The genetic variation of this species was done using DNA sequences from four selected states: Ondo, Osun, Ekiti and Oyo States. Fresh leaves of Gambeya albida were collected from the selected States. The leaves were dried using desiccator containing Calcium Chloride, grinded into powdering form. Followed by DNA isolation in the laboratory, using CTAB Protocol. Polymerase Chain Reaction (PCR) was done using RcbL primers, and deoxyribonucleic acid (DNA) sequence were done using PCR machine and sequencer. The results from the sequencer were analyzed for the final results. The molecular characteristics showed barcode length (bp) of 991, number of variable sites of 494 and Parsimony-Informative sites of 323. While the constructed Phylogenetic tree exposed the degree of similarity among the clusters, taxa and taxon of selected samples. The phylogenetic tree result showed that taxa from the cluster of Esure-Ekiti and Gbongban were the oldest while that of Iropora-Ekiti and Eleyowo were the youngest in genetic evolutionary of Southwestern Nigeria. The result obtained from this study showed that there were significant differences among genetic variation in Gambeya albida. Domestication of Gambeya albida is recommended for our farmers and foresters because of high rate of fruits production by Gambeya albida which might boost the economics of Nigeria..
